Bioactive conc. in 10% aq soln comprising as active principle 7-9% of an anti-hyaluronidase & antiinflammatory mucopolysaccharide polymer & 1-3% of chondroitin sulfate; said active principle having yellow to light brown color, pH 4-6 & an antihyaluronidase activity 450-750 I.U./ml & prepd by process - Extracting at 0-4C bovine cartilaginous connective tissue with aq acid soln contg phenol (0.5%) & having pH 2-4 in a first phase in ratio of 1:3.5 to 4 of tissue to aq soln to obtain first extract & in second phase in ratio 1:1.5 of tissue to aq soln to obtain second extract, said first & second phases each taking place for 24 hours, stirring for 30 min every 3 hours - Combining both extracts - Adjusting pH 4.5-5 with Ca hydroxide, allowing to settle for 8 hours at 0-4C - Filtering & concentrating filtrate by vacuum evaporation at < 45C until rate of concentration of 7:1 - Passing extract through anion exchanger column to obtain aq soln - Removing lipids from aq soln by solvent extraction in ratio of 1:4 soln to extraction solvent in 6-10 cycles until lipids re maining in aq soln < 1-5%, the last defatting cycle carried out with n-hexane in same ratio - Vacuum-concentrating aq soln until reaching vol. of 70% of aq soln before said concn. step - Filtering to obtain filtrate & protein ppcc & adding 96 degree EtOH for deproteinization to filtrate until alc. concn of 50% is reached - Maintaining aq. alc. soln at 0-5C for 12 hours, then filtering & concentrating to remove alc. until reaching a vol. of 70% against soln subject to deproteinization - Gradient-treat. protein ppcc with NH4 sulfate to obtain a low MW fraction of protein ppcc & combining low MW protein ppcc with aq soln concentrated in previous step - Removing any histaminic impurities from combined low MW protein ppcc & aq soln by successive treat with tri-Na phosphate, Ca chloride & NaOH until reaching pH 8.5 filtering combined protein ppcc & aq soln & passing filtrate through cationic exchanger column |
