Dynamic kinetic resolution technique by combining the actions of lipase enzyme & Ru catalyst In the kinetic resolution part of the process, the lipase catalyzes acylation of one enantiomer of a racemic alc. much faster than the other enantiomer. The resolution is called "dynamic" because a Ru catalyst continuously racemizes the unwanted enantiomer of the alc., resulting in a theoretical conversion of 100% of the racemic alc. to the ester of the desired enantiomer. For example the Swedish workers use a commercially available immobilized lipase from the "fungus Candida antarctica" to catalyze acetylation of racemic 2-octanol with p-chlorophenyl acetate. The isolated yield of (R)-2-octyl acetate is 80%, with enantiomeric excess > 97%. The ligand for the Ru catalyst is 2 moles of tetraphenylcyclo pentadienol |
