RESEARCH Strategy involved breaking beta-amyloid aggregates. Idea was to inaugurate a molecular competition: NITROPHENOLS have some chemical characteristics similar to beta-amyloids, like the fact of not dissolving easily in water. Being both beta-amyloid & nitrophenol in proximity, it would be possible that beta-
amyloid adhered to nitrophenol instead of adhering to each other, avoiding formation of neurotoxic plaques.
In experiences done with cell cultures, not only beta-amyloid aggregate plaques did not form, but nitrophenol also prevented neuron cell death.
Next challenge was to verify this discovery in vivo. Researchers simulated a Alzheimer-like condition in RODENTS (rats). Each of them gained on both sides of their Hipocamp (region of brain base linked to memory) very damaged by disease) a injection of beta-amyloid, that immediately formed agglomerates.
Whereas one side of animal’s brain continued to suffer the effects od protein fragment, the other received a injection of nitrophenol. The compd once more showed its efficacy: 85% of aggregates were destroyed & toxic effects on neurons disappeared.
Next step is to develop a disease model in transgenic rats. “In transgenics, the plaques form naturally, although slower. “Although efficacy of nitrophenols should be the same”. Remedy for disease, today uncurable will take with luck 5 – 7 years
