Method of prepg biol. tissue for cryopreservation (vitrification - Flushing the tissue with a cryoprotectant-free stabilizing flushing soln., wherein the tissue is treated with TGF beta1 at least one of prior to or during step (a) - Superfusing the cryoprotectant-free tissue of (a) with super fusion soln under controlled conditions in which the concn of cryoprotectant is gradually increased in the superfusing soln to a first predetermined concn, while the T of tissue & the superfusion soln is concurrently reduced from initial T of 0-37C to -13 - +5C - Maintaining the concn of the cryoprotectant for sufficient ti me to permit the approximate osmotic equilibration of tissue to occur - Increasing the concn of cryoprotectant in the superfusion soln., from concn in (c) under controlled conditions to first intermediate concn., which is not sufficient for vitrification & maintaining the concn of cryoprotectant at that intermedia te concn for sufficient time to permit the approximate osmotic equilibration of the tissue - Reducing T of the tissue to T < that of step (b) - Increasing concn of cryoprotectant in superfusion soln., to level sufficient for equilibration |
