Assay for detecting presence of botulinum toxin neutralizing Ab in serum sample by visual identification of Ab/antigen complex - Obtaining serum from patient suspected of having develo ped neutralizing Ab to botulinum toxin - Separating botulinum toxin protein complex which has not been treated with either Na dodecyl sulfate or trypsin by acrylamide gel electrophoresis, said sepn occurring based on protein size & charge - Electrophoretically transferring separated toxin portions onto solid support wherein each separated toxin protein is bound to solid support at spaced apart sites - Blocking all remaining binding sites on solid support - Contacting solid support with patient serum sample - Contacting solid support with an enzyme labeled anti-hu man Ig Ab - Contact solid support with a chromogenic substrate which reacts with said labeling enzyme to produce colored ppcc - Correlating presence of any colored ppcc formed at one or more sites on solid support which contains separated toxin protein to the presence of said botulinum toxin Ab in human serum sample |
