PATENT CLAIMS Endotoxin-induced inflammation assay for evaluating a drug
- Adm a drug to ocular tissue of an experimental animal
- Inducing inflammation in animal by adm endotoxin
- Isolating the ocular tissue 16-30 hours after adm of endoto
xin
- Homogenizing the ocular tissue in a buffered compn com
prising a non-interfering thiol reactive compd, compn with
pH 5-8
- Centrifuging the compn to recover neutrophils (PMN) &
myeloperoxidase (MPO) in a pellet
- Solubilizing the said pellet in a buffered compn contg
hexadecyltrimethylammonium bromide (HTA-Br) in order
to release MPO from PMN azurophil granules, wherein the
compn has pH 5-7
- Centrifuging compn to recover MPO in the supernatant
- Prepg a buffered compn comprising said supernatant,
o-dianisidine (o-DA), H2O2 & HTA-Br, compn has pH 6
- Determn MPO activity by spectrophotometrically monito
ring at 460 nm change in absorbance of compn due to
o-DA oxidation
