A method for processing a protein, non-protein amino acid polymer or peptide having crosslinked structure - Contacting glutamine & lysine residues in a protein etc., with a transglutaminase obtained from "Bacillus subtilis" to form intermolecular or intramolecular, crosslinked epsilon- (gamma-Glu)-Lys bonds between or in the molecules of the protein etc Wherein said transglutaminase has the following physicoche mical properties - It is active at pH = 7-9 - It is active at 40-65C - It is stable < 60C - The enzymatic activity is not dependent on Ca ion & has an activity > 50% in presence of 5 mM of Ca ion - Inhibited by N-Et maleimide, cystamine or NH4 sulfate - Not inhibited by EDTA, dithiothreitol or HS-CH2-OH - Has MW of (i) 18,000-22,000 as measured by gel permea tion, & (ii) 28,000-30,000 measured by SDS-PAGE - Catalyzes the transacylation of the gamma-carboxyamide group in glutamine residue(s) in peptide chain |
