Method for processing a protein, a non-protein amino acid polymer, or peptide having crosslinked structure - Contacting glutamine & lysine residues in protein etc with transglutaminase obtained from "Bacillus subtilis" to form intermolecular or intramolecular, crosslinked epsilon-(ga mma-Glu)-Lys bonds between or in the molecules of the protein etc Wherein said transglutaminase has the following physicochemical properties - It is active pH = 7-9 - It is active between 40-65C - It is stable at < 60C - The enzymatic activity of the transglutaminase is not depen dent on Ca ion & has activity of > 50% in the presence of 5 mM of Ca ion - It is inhibited by N-Et maleimide, cystamine or NH4 sulfate - It is not inhibited by EDTA, dithiothreitol or 2-mercapto EtOH - It has MW 18,000-22,000 as measured by gel permeation & 28,000-30,000 as measured by SDS-PAGE - It catalyzes the transacylation of the gamma-carboxyamide group in glutamine residue(s) in peptide chain |
