PATENT CLAIMS |
An intact deglycosylated lactoferrin protein or a single domain, deglycosylated lactoferrin protein fragment produced by a process that comprises culturing a transformed Aspergillus fungal cell contg a recombinant plasmid, wherein said plasmid comprises the following comp operably linked from 5' to 3' - Promoter - Nucleotide seq encoding signal peptide - A 5' portion of nucleotide seq of a gene encoding an amino terminal portion of a highly expressed endogenous, secre ted Aspergillus protein - Nucleotide seq encoding peptide linker, said linker com prising cleavage site of protease endogenous to Aspergi llus - Nucleotide seq. encoding lactoferrin or lactoferrin protein fragment Wherein said transformed Aspergillus fungal cell is cultured in a suitable nutrient medium until a lactoferrin protein or fragment thereof is produced as a fusion prodt & then processed via an endogenous proteolytic enzyme specific for said linker seq., wherein said processed lactoferrrin protein or fragment thereof is secreted into the nutrient medium & isolated therefrom & wherein the lactoferrin protein or fragment thereof has been deglycosylated |
