Claim 1 An apparatus for seeding & culturing replacement cartilage tissue constructs within fluid media - At least one wall defining a seeding & culturing chamber - A porous substrate disposed within said chamber configu red & dimensioned to seal with the chamber, said substrate designed to facilitate threee-dimensional tissue growth on said substrate & comprising a three-dimensional frame work having interstitial spaces bridgeable by cells - Means for alternatingly creating a pressure differential within the chamber across the substrate to force fluid within the chamber to flow through the substrate - A support structure located within said chamber configured & dimensioned to support the substrate such that the subs trate is sealed with the chamber to direct all fluid flow within the chamber through the substrate Claim 24 Method for seeding & culturing replacement cartilage tissue constructs in a chamber defined by at least one wall, comprising - Disposing within the chamber a porous substrate configu red & dimensioned to seal with the chamber, said substrate designed to facilitate three-dimensional tissue growth on said substrate & comprising a three-dimensional frame work having interstitial spaces bridgeable by cells - Exposing the substrate in the chamber to a flow of fluid media for seeding & culturing - Alternatingly creating a pressure differential across the substrate during seeding & culturing to force all flow of the fluid media within the chamber through the substrate & faci litate exposure of substrate to the fluid media |
