Molecules that bind to a protein target are identified by using NMR to detect binding-induced chemical shifts changes in the protein. The molecules so identified typically bind to the protein with low affinity, but they are then combined in pairs to create compds with higher affinity. The technique requires repeated screening of large compd libraries & a large amount of a labeled protein target. An advantage of technique is that it !gives structural information about the ligand-protein interaction". Another advantage is that a larger library is screened, increasing the chances of discovering useful lead molecules |
