Process of purifying & recovering recombinant beta-casein from a cell - Extracting said beta-casein from concd. suspension of cells using a water-miscible organic solvent selected from: ace tone, Iso-PrOH, EtOH & MeOH, said solvent contg 50-95% organic solvent & 50-5% water at pH 6.5-8.5 & at 30-50C molar ratio of Cell suspension/Solvent = 1:1 - 1:5 - Isolating beta-casein from cell suspension - Concentrating recombinant beta-casein to remove solvent - Cooling to 5C to separate solids - Isolating beta-casein from solids - Treat. beta-casein with water-soluble Ca salt; Ca salt/Ca sein = 1:1 - 1:5 - Precipitating beta-casein using an acid - Washing with NaOAc - Recovering recombinant beta-casein |
