C) determn comprising
- Diluting whole blood sample with a diluent to give a diluted
whole blood test sample, said diluent consisting of: Na sul
fate, NaCl, EDTA, imidazole, & mixt of hexahydro-1,3,5-tris
(2-hydroxyethyl)-s-triazine & Na 2-pyridinethiol-1-oxide
- Mixing said diluted blood test sample with an aq multipurpo
se lyse reagent to give a whole blood sample/lyse rea
gent mixt., said mixt forming a chromogen, said reagent
comprising 0.1-20% quat ammonium salt, said at least one
quat ammonium salt selected from: tetradecyltrimethyl
ammonium bromide, dodecyltrimethyl ammonium chloride
cetyltrimethyl ammonium bromide, hexadecyltrimethyl
ammonium bromide, benzalkonium chloride & cetylpyridium
chloride & 0.1-15% of hydroxylamine salt selected from:
hydrochloride, sulfate, phosphate & other acid salts
- Presenting said whole blood sample/lyse reagent mixt
to an absorbance spectrophotometer
- Measuring the optical d. of the chromogen
- Determining total Hb concn in sample from the measured
optical d. of the chromogen, wherein the chromogen has
max absorbance of 530-550 nm
- Presenting said whole blood sample/lyse reagent mixt to
particle analyzing system
- Detecting WBC populations
- Determining the number of white blood cells in said WBC
populations in said whole blood sample
