The researchers have designed MULTIVALENT LIGANDS that have the potential to organize cellular receptors to amplify their response to a chemical signal. For example GALACTOSE elicits a CHEMOTACTIC response in E. coli bacteria - that is, the bacteria move towards higher concn of the sugar. Using ring-opening metathesis polymn, the group synthesized 10-, 25-, & 50- mers of galactose & characterized the response of E. coli to them The 10-mer ligand is as effective as galactose itself. Modeling studies indicate that the max length of this polymer is 50 angstroem, making it unlikely that it could cluster galactose R that have been shown to be 90 angstroem apart. The 25-mer & 50-mer are much more effective in eliciting chemotaxis than galactose, with the 25-mer stimulating the behavior at 100-fold lower concn than required for galactose & 50-mer acting at 5,000-fold lower concn |
