The present approach; incorrectly folded proteins are REFOLDED by small-mole. enzyme mimic. When multiple disulfide contg proteins are expressed in yeast, many form nonnative disulfide bonds that prevent them from folding properly. Disulfide cleavage & re-formation is catalyzed in cells by PDI enzyme
The researchers synthesized BMC (trans-1,2-bis(mercapto-
acetamido)cyclohexane (see structure) & tested. They found that it catalyzes disulfide bond isomerization both in vitro & in vivo
