The enzyme is involved in - Catalysis of Carcinogenic Epoxides to harmless 1,2-diols - Catalysis of lipid epoxide leukotoxin into a diol involved in ARDS inflammatory condition So structure determn was important Previous studies, the researchers had good idea of catalytic triad that attacks the epoxide. The X-ray structure confirmed this triad & revealed a key role played by 2 active-site tyrosines in activating the epoxides for attack. The structure also showed that the dimeric enzyme contains 2 types of active sites. An active site for epoxide hydrolysis is found in each of the enzyme's 2 catalytic domains. A second active site, located in the hydrolase's "vestigial" domains, is homologous to the active site of a completely different enzyme, a haloacid dehalogenase. This second site is apparently inactive |
